Abstract:This study was carried out with the purpose to investigate the effect of different doses of sildenafil citrate (Viagra, Revatio) on the levels of phosphodiesterase-5 (PDE5) enzymes and plasma concentrations of sildenafil citrate in men with erectile dysfunction. The study was carried out with a total of 12 men with erectile dysfunction and one with normal erectile function.
Methods:A prospective observational study was carried out with a total of 120 men, who were aged 65 years and over who had erectile dysfunction. The subjects were randomly selected from the general population in the general public (men between the ages of 18 and 41 years, and women between the age of 45 and 59 years). The study was conducted on the basis of the criteria proposed by the International Index of Erectile Function and the Internationalrection Quality Index (IIEF) (WHO). The men were followed up at the same time of day with the same examinations and measurements of blood pressure (BP). The study was approved by the Research Ethics Committee of the Faculty of Medicine and Pharmacy (Institutional Animal Ethics Committee, Ministry of Health, Thailand). Informed consent was obtained from all the participants before the study. The patients with a male/female ratio of more than two were excluded from the study.
Results:The mean age of the patients was 58 years, and the male/female ratio was more than three (53.2%). The average age of the men was 50 years, and the average age of the female was 49 years. The patients were followed up at the same time of day for the same 24 h intervals. There was a positive correlation between blood pressure and blood concentration of sildenafil citrate, but no significant correlation was found between the sildenafil citrate concentrations and age.
Conclusion:In this study, the levels of phosphodiesterase-5 (PDE5) enzymes and plasma concentrations of sildenafil citrate were higher in men with erectile dysfunction, but no significant difference was found in plasma concentrations of sildenafil citrate.
Viagra and Revatio: a clinical study.IntroductionViagra is an effective treatment for erectile dysfunction in men. The mechanism of action of sildenafil, the active ingredient of Viagra, has been previously described in several studies. Sildenafil is a phosphodiesterase (PDE) inhibitor. It works by inhibiting cyclic guanosine monophosphate (cGMP) production in the corpus cavernosum of the penis. The action of sildenafil is due to inhibition of the degradation of cGMP in the corpus cavernosum, and the inhibition of phosphodiesterase type 5 (PDE5) is an action of sildenafil.
MethodSildenafil was purchased from a pharmaceutical company and mixed with 1 mg/ml of sodium phosphate anhydrous. The concentration of sildenafil was 100 mg/l. The amount of the drug was measured at the same time of day. The sildenafil citrate and sildenafil were administered as a single dose. The drug was dissolved in 50% dimethyl sulfoxide (DMSO) and a pH of 7.2 was adjusted to pH of 7.4 using NaOH.
